Technology to Find one Valuable and Highly Functional
Cell from the Rotational Speed of the Cell Without
Labeling

When cells are exposed to a rotating electric field, they polarize and rotate. Rotation rate reflects the roughness of the cell surface, thus the state and type of cells can be identified without labeling. Cells are rotated by an electrode device with a unique microwell structure. Since cells are retained in the wells, their rotation rates are monitored during stimulation with chemicals and antigens. Valuable and highly functional cells can be identified intact based on their rotation rate to these stimulants. The identified cells are collected and cultured to provide a highly functional and homogeneous cell population.

For example, in the fields of drug discovery, drug screening, and medicine, cells that secrete antibodies as molecular target drugs, as well as cells that attack cancer cells, have been utilized as source cells for the production of antibodies or cell therapy. However, the level of cell function varies from cell to cell, even within the same cell type. Even if highly functional cells were to be identified, it would have been impossible to use them because labeling and disruption are required to evaluate cell function. In this research, highly functional cells can be easily and promptly identified, retrieved, and cultured using only cell rotation rates, enabling efficient drug discovery, drug screening, and homogeneous cell medicine using highly functional cell populations.

Highly Functional Cell Populations Can Be Found and Acquired in a Perfectly Usable State

Our unique technology is an electrode device that combines microwells that can monitor the function of individual cells in a time series manner with an electrorotation method that can evaluate the roughness of the cell surface layer. This electrode device enables the evaluation of cell function by measuring the rotation rates of individual cells without labeling. In addition, single cells can be retrieved and cultured from the device according to the rotation rates to obtain a highly functional and homogeneous cell population.

With the spread of genome editing technology and the development of systems biology, it has become easier to artificially modify and enhance cellular functions. Such cells are called designer cells and are expected to be applied to cell therapy and material production. Our vision is to expand their potential by making designer cells more accessible by incorporating this technology into the quality control process of designer cells to realize low-cost cell-based therapy and manufacturing.
